ABSTRACT

Objective: to investigate the effect of two natural cross-linkers on microtensile bond strength (µTBS) and evaluate their influence on the durability of the resin dentin bonds. Material and Methods: the Moringa oleifera and Centella asiatica plant extracts were qualitatively tested with high-performance thin layer chromatography (HPTLC) for the presence of phenols. The phenolic content ranged from 27 to 30 gallic acid equivalents (GAE), µg/mg of dry weight. After etching, two concentrations (5% and 1%) of these two extracts were prepared and used as pretreatment liners on dentin. They were applied for a min. After restoration with resin composite, dentin resin beams were prepared. The study groups were 5% Moringa, 1% Moringa 5% Centella 1% Centella, and control (without cross-linker application). For each group, half of the samples underwent µTBS testing after 24 hours, while the remaining half were immersed in artificial saliva to assess the bond's longevity after 6 months of ageing. Statistical analysis was performed using one-way ANOVA followed by Tukey's post hoc test. Results: both 5% and 1% Moringa showed a significant difference (p<0.05) compared to the other groups at both intervals. However, after ageing, the specimens in the control and 1% Centella groups resulted in a significant decrease in µTBS. Conclusion: overall, both concentrations of Moringa (5% and 1%) were effective in stabilising the bond during both intervals.(AU)


Objetivo: investigar o efeito de dois reticuladores naturais na resistência de união (µTBS) à microtração e avaliar sua influência na durabilidade da adesão da resina à dentina. Material e Métodos: extratos das plantas Moringa oleifera e Centella asiatica foram qualitativamente testados através de cromatografia em camada fina de alta performance (HPTLC) para a presença de fenóis. O conteúdo fenólico alcançou entre 27 a 30 equivalentes de ácido gálico (GAE), µg/mg de peso seco. Após o condicionamento, duas concentrações (5% e 1%) dos extratos foram preparadas e utilizadas como forros de pré-tratamento em dentina. Eles foram aplicados por um minuto. Após a restauração com resina composta, palitos de dentina e resina foram preparados. Os grupos foram 5% Moringa, 1% Moringa, 5% Centella, 1% Centella e controle (sem aplicação de reticulador). Para cada grupo, metade das amostras foram submetidas ao teste µTBS após 24 horas, enquanto a outra metade foi imersa em saliva artificial para avaliar a longevidade adesiva após 6 meses de envelhecimento. Foi realizada análise estatística através de ANOVA 1-fator, seguido do teste post hoc de Tukey. Resultados: ambas as concentrações de 5% e 1% de Moringa demonstraram diferença significativa (p<0.05) comparadas aos outros grupos em ambos os intervalos. No entanto, após o envelhecimento, os espécimes dos geupos controle e 1% de Centella resultaram em uma redução significativa de µTBS. Conclusão: no geral, ambas as concentrações de Moringa (5% e 1%) foram efetivas em estabelecer a adesão em ambos os intervalos (AU)

ABSTRACT

Low water potential related stresses are regulated by modifying water uptake and loss to avoid low water potential, accumulating solutes which in turn enhance active principles and its gene expressions. Present study examined effect of in vitro induced absorption of mannitol and PEG (poly ethelene glycol) 6000 in Indian pennywort, Centella asiatica (L.) Urb., neutraceutical plant, evidenced by phenotypic, molecular and phytochemical analyses. Both mannitol and PEG 6000 induce water deficit conditions in plants and retarded normal plant biomass in terms of fresh and dry weights. These effects were significantly less severe in plants subjected to mannitol, compared to PEG. PEG and mannitol imposed water deficit, resulted in decline in major active compound, asiaticoside evidenced by HPTLC of asiaticoside content. Differential expression of some selected key genes in the asiaticoside pathway including squalene synthase and ? amyrin synthase by qPCR, confirmed decrease in transcript level expression of asiaticoside, whereas upregulated transcript level expression was observed in cycloartenol synthase for synthesis of phytosterols. Estimation of total flavonoids and phenolics under different water deficit conditions were found declined. In conclusion, water deficit by mannitol and PEG 6000 can significantly affects processes associated with biomass growth and ability to synthesize secondary metabolites in C. Asiatica.

Objective:This study aimed to determinate ten phenols (polydatin, resveratrol, rhein, emodin, chrysophanol, physcion, oxyresveratrol, 2,3,5,4'-tetera-hydroxystilbene-2-O-β-D-glucoside, (+)-catechin and (-)-epicatechin) in Polygoni Cuspidati Rhizoma simultaneously based on the high-resolution multiple reaction monitoring (MRMHR) mode of ultra- performance liquid chromatography- quadrupole/time-of-flight mass spectrometry. Methods:The assay was performed on Waters ACQUITY UPLC BEH C18 (2.1 mm× 100 mm, 1.7 μm) column using acetonitrile-0.1% formic acid as the mobile phase. The flow rate was 0.2 ml/min. The MRMHR mode was adopted for quantification.Results:The analyzed compounds showed good linearity relationships ( r2>0.999). The intra- and inter-batch relative standard deviations were all <5% and the recovery rate was between 96.28%-103.23%. The content of polydatin was the highest, followed by resveratrol and emodin. However, the contents of chrysophanol and oxyresveratrol were relatively low and some batches were unqualified. The contents of analyzed compounds varied significantly among the ten batches. Conclusion:The proposed UPLC-Q/TOF-MS method was successfully established to determinate ten phenols in Polygoni Cuspidati Rhizoma simultaneously, which could provide technical support for the quality evaluation of Polygonum cuspidatum.

Abstract The objective of this work was to perform the phytochemical characterization, to determine total phenols, antioxidant (AAO%) and antimicrobial potential of the ethanolic extracts of carambola. The phytochemical study was carried out through a qualitative analysis of the chemical constituents and quantitative determination of the phenol content By the Folin-Ciocalteu test. Qualitative and quantitative antioxidant tests were performed using the DPPH method (2,2 diphenyl-1-picryl-hydrazila) and iron reduction (FRAP). The minimum inhibitory concentration (MIC) was determined by microdilution in 96-well plates. The presence of pyrogallic tannins, steroids and saponins has been identified. The highest total phenol content, quantified in the samples, was found in the stem bark (0.0866 mgEAG/g) and in the fruit (0.0734 mgEAG/g). In the antioxidant evaluation, the extracts of the green fruit bagasse (AAO% 71.9%,) and stem bark at 50 μg/mL (AAO% 94%) with CE50 23.7 μg/mL. Leaf extracts, stem bark, ripe fruit bagasse and green fruit bagasse presented MICs of 100 μg/mL against multiresistant pathogenic bacteria and fungi.


Resumo O objetivo desse trabalho foi realizar a caracterização fitoquímica, determinar fenóis totais, potencial antioxidante (AAO%) e antimicrobiano dos extratos etanólicos de carambola O estudo fitoquímico foi realizado por meio de análise qualitativa dos constituintes químicos e determinação quantitativa do teor de fenóis totais pelo teste de Folin-Ciocalteu. Os testes antioxidantes qualitativos e quantitativos foram realizados pelo método do DPPH (2,2 difenil-1- picril-hidrazila) e redução do ferro (FRAP). A concentração inibitória mínima (CIM) foi determinada por microdiluição em placas de 96 poços. Foi identificada a presença de taninos pirogálicos, esteroides e saponinas. O maior teor de fenóis totais, quantificado nas amostras, foi encontrado na casca do caule (0,0866 mg EAG/g) e no fruto (0,0734 mg EAG/g). Na avaliação antioxidante destacaram-se a 500 µg/mL os extratos do bagaço do fruto verde (AAO% 71,9%,), e casca do caule a 50 µg/mL (AAO% 94%) com CE50 23,7 µg/mL. Os extratos das folhas, casca do caule, bagaço do fruto maduro e bagaço do fruto verde apresentaram CIM de 100 µg/mL contra bactérias e fungos patogênicos multirresistentes.

ABSTRACT

Background: Asparagus (Asparagus officinalis L.) green is a vegetable with a great demand worldwide, and likewise, it is highly perishable, due to its high respiration rate that accelerates its senescence. Disinfection of vegetables after their harvest is an obligatory practice that can reduce losses by decomposition due to the attack of microorganisms. Therefore, it is vital to preserving its microbiological and sensory characteristics to reach the final consumer. Objective: to evaluate the effect of gaseous ozone (0 to 10 ppm) and storage time (0 to 30 days) on phenol content, overall appearance, count of molds, psychrophilic bacteria, and viable mesophilic aerobes. Methods: the response surface methodology was used, applying a rotatable central composite design. Results: the results indicated that there was a significant influence (p <0.05) of the independent variables on the characteristics studied, as well as an adequate lack of fit of the quadratic regression model (p> 0.05). By means of the contour superposition technique, it was determined that the optimal conditions for the highest retention of phenol content (16.99 mg/g) and overall appearance (7.61 points) and lower counts of viable aerobic mesophilic bacteria (5.3 x 103 CFU/g) they corresponded to 10 ppm of gaseous ozone up to 25.91 days of storage, with adequate quality characteristics in the spears. Conclusion: the region of interest was determined for optimal retention of phenol content and overall appearance, and a lower count of viable aerobic mesophilic bacteria in green asparagus during postharvest, suggesting to use the initial application of ozone gas at 10 ppm allowing 25.9 days storage at 1 °C. The results indicate that this technology is a good alternative in the conservation of fresh vegetables


Antecedentes: El espárrago (Asparagus officinalis L.) verde; es una hortaliza con una gran demanda a nivel mundial, y, asimismo, es altamente perecible, por su elevada velocidad de respiración que, acelera su proceso de senescencia. La desinfección de los vegetales después de su cosecha es una práctica obligada que puede disminuir las pérdidas por descomposición debido al ataque de microrganismos. Por lo tanto, es muy importante conservar sus características microbiológicas y sensoriales para llegar al consumidor final. Objetivo: evaluar el efecto del ozono gaseoso (0 a 10 ppm) y tiempo de almacenamiento (0 a 30 días) sobre el contenido de fenoles, apariencia general, recuento de mohos, bacterias psicrófilas y aerobias mesófilas viables. Métodos: se utilizó la metodología de superficie de respuesta, aplicando un diseño compuesto central rotable. Resultados: los resultados indicaron que existió influencia significativa (p<0.05) de las variables independientes sobre las características estudiadas, así como, una adecuada bondad de ajuste del modelo de regresión cuadrático (p>0.05). Mediante la técnica de superposición de contornos se determinó que las condiciones óptimas para la mayor retención de contenido de fenoles (16.99 mg/g) y apariencia general (7.61 puntos) y menor recuentos de bacterias aerobias mesófilas viables (5.3 x 103 UFC/g) correspondieron a 10 ppm de ozono gaseoso hasta los 25.91 días de almacenamiento, con adecuadas características de calidad en los turiones. Conclusión: se determinó la región de interés para una óptima retención de contenido de fenoles y apariencia general, así como, menor recuento de bacterias aerobias mesófilas viables en el espárrago verde durante la postcosecha, sugiriendo utilizar la aplicación inicial de ozono gaseoso a 10 ppm permitiendo 25.9 días de almacenamiento a 1 °C. Los resultados indican que esta tecnología es una buena alternativa en la conservación de hortalizas frescas

ABSTRACT

Objetivo: Determinar el contenido de compuestos bioactivos y la capacidad antioxidante de los frutos tumbo (Passiflora mollissima) y cerezo (Prunus serotina). Materiales y métodos: Estudio analítico, experimental, longitudinal y prospectivo. Los frutos cerezo y tumbo se recolectaron en las regiones de Cusco, Moquegua y Arequipa. La técnica de Folin-Ciocalteu fue empleada para determinar el contenido de fenoles, y el cloruro de aluminio se utilizó para calcular los flavonoides. La actividad antioxidante se evaluó mediante las técnicas Ferric Reducing Ability of Plasma (FRAP), 2,2-difenil-picril-hidrazil (DPPH) y sustancias reactivas al ácido tiobarbitúrico (TBARS). Para estudiar el efecto hepatoprotector de las frutas, utilizamos ratas albinas que se clasificaron en un grupo control negativo, un grupo control positivo y cuatro grupos experimentales. Resultados: La mayor concentración de fenoles totales y flavonoides se encontró en el tumbo de la región Cusco (Quechua) (fenoles totales: 584,94 ± 134,62 mg EAG / 100 g y flavonoides :445,62 ± 7,94 mg EQ / 100 g). Para el radical DPPH, el valor IC50 del tumbo de la región Arequipa (Yunga) fue 0,41 ± 0,01 mg / mL. El tumbo de la región Cusco (Quechua) mostró el valor FRAP más alto (8,38 ± 0,32 mmol Fe2 + / 100 g). El cerezo de la región de Arequipa (Yunga) presentó la mayor concentración de fenoles totales (181,81 ± 34,1 mg EAG / 100 g) y flavonoides (205,18 ± 77,8 mg EQ / 100 g). El cerezo de Arequipa (Yunga) mostró una actividad antioxidante significativa al DPPH (2,1 ± 0,01 mg / mL), mientras que la capacidad antioxidante del cerezo de la región Cusco (Quechua), evaluada con la técnica FRAP, alcanzó un valor de 1,59 ± 0,2 mmol Fe2+/100 g. Las diferencias observadas fueron estadísticamente significativas. El tumbo mostró un mejor efecto hepatoprotector que el cerezo. Conclusiones: El tumbo de la región Cusco (Quechua) es una fuente importante de compuestos antioxidantes y muestra una elevada capacidad antioxidante (FRAP), mientras que el cerezo de la región Arequipa (Yunga) tiene un alto contenido de compuestos antioxidantes y una mayor capacidad antioxidante (DPPH).


Objective: To determine the content of bioactive compounds and antioxidant capacity of the banana passionfruit (Passiflora mollissima) and black cherry (Prunus serotina). Materials and methods: An analytical, experimental, longitudinal and prospective study. The black cherries and banana passionfruits were collected in the Cusco, Moquegua and Arequipa regions. The content of phenols and flavonoids were determined using the Folin-Ciocalteu reagent and aluminum chloride method, respectively. The antioxidant activity was evaluated using the ferric reducing ability of plasma (FRAP), 2,2-diphenyl-1-picryl-hydrazyl (DPPH) and thiobarbituric acid reactive substance (TBARS) techniques. Albino rats classified into a negative control group, a positive control group and four experimental groups were used to study the hepatoprotective effect of the fruits. Results: Banana passionfruits from the Cusco region (Quechua) showed the highest concentration of total phenols (584.94 ± 134.62 mg GAE/100 g) and flavonoids (445.62 ± 7.94 mg QE/100 g). Concerning the DPPH radical, the IC 50 value of banana passionfruits from the Arequipa region (Yunga) was found to be 0.41 ± 0.01 mg/mL. Banana passionfruits from the Cusco region (Quechua) showed the highest FRAP value (8.38 ± 0.32 mmol Fe2+/100 g). Black cherries from the Arequipa region (Yunga) had the highest concentration of total phenols (181.81 ± 34.1 mg GAE/100 g) and flavonoids (205.18 ± 77.8 mg QE/100 g). They also showed a significant antioxidant activity regarding the DPPH (2.1 ± 0.01 mg/mL), while the antioxidant capacity of black cherries from the Cusco region (Quechua), which was evaluated with the FRAP method, achieved a value of 1.59 ± 0.2 mmol Fe2+/100 g. The observed differences were statistically significant. Banana passionfruits showed a better hepatoprotective effect than black cherries. Conclusions: Banana passionfruits from the Cusco region (Quechua) are an important source of antioxidant compounds and show a high antioxidant capacity (FRAP), while black cherries from the Arequipa region (Yunga) have a high content of antioxidant compounds and a higher antioxidant capacity (DPPH).

Abstract Introduction: The organoleptic qualities of aromatic species and their derived products are directly related to some characteristics of flavor, color and nutritional value and depend largely on their genetic origin and content of secondary metabolites. Objective: The antioxidant activity of different genotypes of Theobroma spp. from Mexico was evaluated in order to distinguish promising qualities for genetic improvement, and to differentiate phylogenetic traits, considering biochemical variables. Methods: The amount of phenols, flavonoids and antioxidant activity was determined by ABTS and DDPH, in addition to the content of anthocyanins, theobromine and caffeine in four species of Theobroma L., and 50 genotypes derived from T. cacao. The results were analyzed using an analysis of variance, means test, principal component analysis and cladistic analysis. Results: There are highly significant differences between genotypes. The phenol content ranged from 7.5-85 mg g-1; flavonoids 6.57-69.6 mg g-1, antioxidant activity by ABTS of 17.3-86.1 and by DDPH of 40.0-53.3; anthocyanin content of 0.01-3, caffeine of 1.8-6.7-and theobromine of 2.9-9.8 mg g-1. Principal component and cladistic analysis helped explain the variation found and distinguish evolutionary characters and phylogenetic brotherhoods. The variation in content of phenols, flavonoids, antioxidant activity, anthocyanins, theobromine and caffeine was mainly due to the degree of domestication, while for the group of genotypes derived from T. cacao (forastero, trinitario and criollo) it was the origin of the seeds. Conclusions: The degree of domestication influences the content of phenols and antioxidant capacity. The results suggest that the evaluated variables can help to form criteria for genetic improvement in the complex derived from T. cacao oriented to the selection of higher phenol content and greater antioxidant activity.


Resumen Introducción: Las cualidades organolépticas de las especies aromáticas y sus productos derivados se relacionan directamente con algunas características del sabor, color y valor nutricional y dependen en gran medida de su origen genético y contenido de metabolitos secundarios. Objetivo: Se evaluó la actividad antioxidante de diferentes genotipos de Theobroma spp. de México, con el fin de distinguir cualidades promisorias para el mejoramiento genético, y diferenciar rasgos filogenéticos, considerando variables bioquímicas. Métodos: Se determinó la cantidad de fenoles, flavonoides y actividad antioxidante mediante ABTS y DDPH, además de contenido de antocianinas, teobromina y cafeína en cuatro especies de Theobroma L., y 50 genotipos derivados de T. cacao. Resultados: Los resultados fueron analizados mediante un análisis de varianza, prueba de medias, análisis de componentes principales y análisis cladístico. Existen diferencias altamente significativas entre genotipos. El contenido de fenoles varió de 7.5-85 mg g-1; flavonoides 6.57-69.6-mg g-1, actividad antioxidante por ABTS de 17.3-86.1 y por DDPH de 40.0-53.3; el contenido de antocianinas de 0.01-3, cafeína de 1.8-6.7 y teobromina de 2.9-9.8 mg g-1. El análisis de componentes principales y cladístico ayudó a explicar la variación encontrada y distinguir caracteres evolutivos y hermandades filogenéticas. La variación en contenido de fenoles, flavonoides, actividad antioxidante, antocianinas, teobromina y cafeína estuvo dada principalmente por el grado de domesticación, mientras que para el grupo de genotipos derivados de T. cacao (forastero, trinitario y criollo) fue el origen de las semillas. Conclusión: El grado de domesticación influye en el contenido de fenoles y actividad antioxidante. Los resultados sugieren que las variables evaluadas pueden ayudar a formar criterios para el mejoramiento genético en el complejo derivado de T. cacao orientado a la selección de mayor contenido de fenoles y mayor actividad antioxidante.

ABSTRACT Beauregard sweet potato is an orange color crop rich in β-carotene, a precursor of vitamin A. β-carotene improve immunity and decrease of incidence of degenerative diseases. The objective of this work was to introduce sweet potato in the diet through of some food products, such as bread, cake and sweets. The effect of processing on the chemical composition, as well as the quantification of phenols and total carotenoids, antioxidant activity, and sensory acceptability were evaluated. Cakes and coconut sweets presented higher levels of carotenoids. Higher phenolic and antioxidant activity were verified in cocoa sweets. All foods had good acceptability: 86% for bread, 83.3% for cake and 84,4%, and 86% for coconut and cocoa sweets, respectively. Servings of 95g of roasted sweet potato, 330g of bread, 182g of cake, 187g of coconut sweet and 391g end of cocoa sweet, would provide the recommended daily intake of vitamin A for children 10 years and older. It was concluded that the elaboration of foods containing sweet potato is a viable alternative to inserting biofortified foods into the human diet.


RESUMEN Los camote Beauregard son genotipos de color naranja, ricos en β-caroteno, precursores de la vitamina A. El β-caroteno ayuda a mejorar la inmunidad y reducir el riesgo de enfermedades degenerativas. El objetivo del trabajo fue introducir el camote en la dieta a través del pan, pasteles y dulces tipo postre. Se evaluó el efecto del procesamiento sobre la composición química, así como la cuantificación de fenoles y carotenoides totales, actividad antioxidante y aceptabilidad sensorial. Las tortas y caramelos de coco mostraron mayores niveles de carotenoides y cacao, mayor actividad fenólica y antioxidante. Se confirmó una buena aceptabilidad, en la cual el pan presentó 86%, el torta 83,3%, y los dulces tipo postre de coco y de cacao 84,4% y 86% respectivamente. Las porciones de 95 g de batatas al horno, 330 g de pan, 182 g de torta, 187 g y 391 g de dulce de coco y dulce de cacao respectivamente, proporcionarían la ingesta diaria recomendada de vitamina A para niños de 10 años en adelante. Se concluyó que la preparación de alimentos que contienen batatas es una alternativa viable a la inclusión de alimentos biofortificados en la dieta humana.

ABSTRACT

The present work describes the in vitro biological activities of the crude extracts (petroleum ether, ethyl acetate and n-butanol) prepared from the species Ephedra altissima Desf. The estimation of total phenolic, flavonoid and tannin contents were carried out using the Folin-Ciocalteu, trichloroaluminum and acidified vanillin methods, respectively. The evaluation of the in vitro antioxidant activities were performed by three different methods namely: scavenging of the free radical ABTS, permanganate reducing antioxidant capacity, and potentiometric assay. In addition, the antibacterial activity was assessed by the agar disk diffusion assay against seven bacterial strains. The results of the phytochemical screening revealed the presence of several types of secondary metabolites. The EtOAc extract exhibited the highest content of phenols (125.62 ± 1.51 & 956;g EGA mg-1 of extract). The greatest flavonoid and tannin contents were observed for n-BuOH extract (19.18 ± 0.39 µg EQ mg-1 of extract and 8.95 ± 1.70 & 956;g EC mg-1 of extract, respectively). Moreover, the EtOAc extract revealed potent antioxidant activity in all the tested methods. Furthermore, the aqueous extract from the species E. altissima showed a good ability to reduce iron III to iron II with a value of 0.68 ± 0.3 moL eq L-1 in potentiometric assay. All the crude extracts (PE, EtOAc and n-BuOH) displayed inhibition of bacterial growth against at least three strains with values of MIC ranging from 3.125 to 50 µg mL-1. Therefore, these results suggest that Ephedra altissima could be used as an important source of natural bioactive compounds with antioxidant and antibacterial properties.

ABSTRACT

Six new bisabolane-type phenolic sesquiterpenoids, including plakordiols A-D (1-4), (7R, 10R)-hydroxycurcudiol (5) and (7R, 10S)-hydroxycurcudiol (6) were isolated from the marine sponge Plakortis simplex collected from the South China Sea. Their structures were determined based on extensive analysis of spectroscopic data. Their configurations were assigned by coupling constant analysis, NOESY correlations, and the modified Mosher's method. Furthermore, their cytotoxic and antibacterial activities were evaluated.

ABSTRACT

This study intends to develop a high performance liquid chromatography-diode array detection(HPLC-DAD) method for simultaneous determination of chlorogenic acid, 2-hydroxymethyl-3-hydroxyl-1-butene-4-O-β-D-(6″-O-caffeoyl)-glucopyranoside, pubescenoside B, huazhongilexone-7-O-β-D-glucopyranoside, rutin, isochlorogenic acid B, isochlorogenic acid A, isochlorogenic acid C in Ilex hainanensis. The HPLC conditions are as follows: Waters XBridge C_(18 )column(4.6 mm×250 mm, 5 μm), mobile phase of 0.5% formic acid in water(A)-acetonitrile(B), gradient elution(0-8 min, 5%-12% B; 8-18 min, 12%-18% B; 18-30 min, 18%-25% B; 30-40 min, 25%-30% B; 40-42 min, 30%-80% B; 42-45 min, 80% B) at the flow rate of 0.8 mL·min~(-1), detection wavelengths of 282, 324, and 360 nm, column temperature of 25 ℃, and injection volume of 5 μL. The content of the 8 phenols in 8 samples was 0.30-6.29, 0.29-3.27, 0.15-10.4, 0.51-5.85, 0.49-9.02, 0.51-4.68, 1.93-13.4, and 0.87-5.95 mg·g~(-1), respectively. Moreover, the content of phenols in the samples collected in October was higher than that of samples harvested in other months. The established method is accurate and sensitive for the determination of phenols in I. hainanensis, which is useful for the quality improvement of this herbal medicine.

ABSTRACT

Objective:To investigate the antioxidant activity and chemical composition of 75% ethanol extract of <italic>Rosa cymosa</italic> roots and its different polar parts. Method:The 75% ethanol extract of <italic>R. cymosa</italic> roots was divided into dichloromethane, ethyl acetate, <italic>n</italic>-butanol and water parts by organic solvent extraction. <italic>In vitro</italic> antioxidant activity of each fraction was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging assays, as well as ferric reducing antioxidant power (FRAP) test. The contents of total triterpenes, total phenols, total tannins and condensed tannins in each fraction were determined by spectrophotometry. SPSS 24.0 software was used to conduct Pearson correlation analysis between the antioxidant activity of each fraction and the content of the main components, and then the main active fraction and the main active components were determined. The chemical constituents of the active fraction was analyzed by ultra performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS/MS), and the structures of the main chromatographic peaks were predicted. Result:Each fraction of <italic>R. cymosa</italic> roots had certain antioxidant activity, and there was a significant dose-effect relationship within a certain concentration range, but the antioxidant activity of different polar parts was different. In DPPH and ABTS free radical scavenging tests, the antioxidant activity of each fraction and vitamin C (VC, the positive drug) was ranked as ethyl acetate fraction>VC><italic>n</italic>-butanol fraction>ethanol extract>water fraction>dichloromethane fraction. In FRAP test, the activity of ethyl acetate fraction was weaker than that of VC, and the other order was unchanged. The contents of total triterpenes, total phenols, total tannins and condensed tannins in ethyl acetate fraction were 3.81%, 50.33%, 3.32%, and 39.79%, in <italic>n</italic>-butanol fraction were 0.88%, 41.42%, 2.25% and 23.55%, in ethanol extract were 2.90%, 41.95%, 3.43% and 20.14%, in water fraction were 0, 26.80%, 16.90% and 7.57%, and in dichloromethane fraction were 21.23%, 12.90%, 1.59%, and 6.17%, respectively. Correlation analysis results showed that the contents of total phenols and condensed tannins were positively correlated with the antioxidant activity, the contents of total triterpenes were negatively correlated with the antioxidant activity, and the correlation between total tannins and antioxidant activity was not obvious. A total of 26 compounds were identified from the ethyl acetate fraction by UPLC-Q-TOF-MS/MS, including 11 condensed tannins, 4 hydrolysable tannins, 6 triterpenes, 3 flavonoids, 1 benzoic acid derivative and 1 chlorogenic acid analogue. Conclusion:Ethyl acetate fraction is the main antioxidant active site of <italic>R. cymosa</italic> roots, and phenols mainly composed of condensed tannins are the main active components. The results can provide experimental basis for the development of natural antioxidants.

Introducción: A nivel mundial el consumo Hibiscus sabdariffa, ha venido creciendo gracias a su contenido rico en compuestos bioactivos como flavonoides y antocianinas. Por medio de sus cálices ha sido utilizada para prevenir y tratar enfermedades degenerativas como el cáncer, anomalías cardiovasculares e hiperlipidemia. Objetivo: Evaluar el efecto antioxidante de un extracto de H. sabdariffa, por medio del ensayo de oxidación de lipoproteínas de baja densidad (LDL) aisladas de plasma sanguíneo. Métodos: Se emplearon cálices deshidratados obtenidos de un cultivo propio, obteniendo un extracto acuso bajo condiciones similares a la forma de consumo habitual. Se determino el contenido de fenoles totales empleando el método de Folin-Ciocalteu. Caracterizado el extracto se evaluó su actividad antioxidante por el ensayo de oxidación de LDL, obtenida de una muestra sanguínea de tres pacientes sin hiperlipidemia. Resultados: Se evidenció una inhibición de la oxidación de LDL estadísticamente significativa del extracto y su respectivo al control en cada una de las muestras ( p<0,05). Conclusiones: estos resultados respaldan que H. sabdariffa, es una alternativa real en el control de la hiperlipidemia y reducción de la oxidación de las LDL, responsable de la formación de la placa de ateroma a nivel de las arterias. Sin embargo el consumo de esta planta debe hacerse bajo condiciones controlados y guiadas por especialistas a fin de obtener los resultados deseados


Introduction: Worldwide consumption Hibiscus sabdariffa, has been growing thanks to its high content rich in bioactive compounds such as flavonoids and anthocyanins. Through its calyces it has been used to prevent and treat degenerative diseases such as cancer, cardiovascular abnormalities, and hyperlipidemia. Objective: To evaluate the antioxidant effect of an extract of H. sabdariffa, by means of the low-density lipoprotein oxidation (LDL) assay. Methods: Dehydrated chalices obtained from an own culture were used, obtaining an acuso extract under conditions similar to the usual consumption. The total phenol content was determined using the Folin-Ciocalteu method. Characterized the extract, its antioxidant activity was evaluated by the LDL oxidation test, obtained from a blood sample from three patients without hyperlipidemia. Results: A statistically significant inhibition of LDL oxidation of the extract and its respective control was evidenced in each of the samples (p <0.05). Conclusions: these results demonstrate that H. sabdariffa is a real alternative in the control of hyperlipidemia and reduction of oxidation of LDL, responsible for the formation of atheroma plaque at the level of the arteries. However, the consumption of this plant must be done under controlled conditions and guided by specialists in order to obtain the desired results.

Abstract Phenolic compounds are one of the main groups of secondary metabolites in plants and are known for their antioxidant activity. Rosmarinus officinalis L. (rosemary) contains different phenolic compounds including carnosol, carnosic acid and rosmarinic acid. In Cuba, rosemary cultivation is limited because it is difficult to propagate and has a low yield. As a result, it was removed from the Herbal Medicine National Formulary. However, the National Public Health System has a strong interest in rosemary because of its value as a natural antioxidant medicine. Irrigation with water treated with a static magnetic field (SMF) is a possible strategy to increase rosemary yield. This technology has been applied to accelerate plant growth and increase crop quality. The aim of this study was to evaluate the content of phenolic compounds and antioxidant activity in aqueous leaf extracts from plants irrigated with SMF-treated water in comparison to control plants. Significant differences in phenolic content and antioxidant activity were observed between aqueous extracts of control plants and plants irrigated with SMF-treated water. Therefore, irrigation with SMF-treated water is a promising technology to improve the cultivation of rosemary as a raw material to obtain pharmaceutical products with high antioxidant activities.

ABSTRACT

Objective: To explore the regulatory effect of malt alkaloid on prolactin (PRL) secretion in the model rat with postpartum hypogalactia induced by bromocriptine based on dopamine D2 receptor, and determine the active fraction of the malt with galactogogue effect. Methods: The postpartum hypogalactia model was established by intragastric administration of bromocriptine mesylate. After the model was successfully established, all groups were given corresponding drug treatment. The concentration of serum PRL, estradiol (E2) and progesterone (P) in each group was detected by ELISA kits. HE staining was used to observe the pathologic changes of breast tissue. RT-PCR was used to determine the mRNA levels of prolactin receptor (PRLR) and dopamine D2 receptor (DRD2) in pituitary gland of rats. Results: Compared with the control group, the levels of serum PRL, P, and E2 were significantly decreased in the model group as well as the mRNA expression of the pituitary PRL cells. But the mRNA expression of the pituitary DRD2 in the model group was significantly increased compared with the control group. Compared with the model group, the malt total alkaloid significantly increased the volume of mammary lobule and dilated the duct. There was a lot of milk in the duct and acinar in the malt total alkaloid group. Besides, the total alkaloids increased the concentration of serum PRL, P, and E2 and the mRNA expression of the pituitary PRL cells, and decreased the mRNA expression of the pituitary DRD2. Conclusion: The primary the active fraction of malt for galactogogue action is total alkaloids, and its mechanism may be related to promoting PRL secretion, increasing serum PRL receptor level and decreasing the mRNA expression of dopamine D2 receptor.

Foeniculi Fructus is warm and spicy in nature and belongs to the liver, kidney, spleen and stomach meridians with the function of dispersing cold and relieving pain, regulating Qi-flowing and harmonizing stomach, and can be used as medicine and food. This article summarized the historical evolution of processing of Foeniculi Fructus by consulting the relevant books since the Han dynasty, the Chinese Pharmacopoeia as well as the processing norms of provinces and cities, and summarized the chemical compositions and pharmacological effects of Foeniculi Fructus by reviewing literatures at home and abroad, then it found that Foeniculi Fructus mainly contains essential oil, flavonoids, phenols and fatty acids, has certain effects on human visceral system, central nervous system, cardiovascular system, endocrine system, immune system and chemotherapy, including the functions of regulating gastrointestinal function, analgesia, anti-inflammatory, etc. Among them, the main chemical components that play the pharmacodynamic effect are volatile oil, flavonoids and phenols, and the chemical components and pharmacological effects in Foeniculi Fructus before and after processing will have corresponding changes, which can provide reference for the further study of Foeniculi Fructus in processing principle, pharmacodynamic substance basis and pharmacological mechanism.

ABSTRACT

Objective:To investigate constituents containing phenolic hydroxyl or carboxylic acid (excluding diarylheptanoids) from Curcumae Rhizoma and Sparganii Rhizoma herbal pair and single herb. Method:Multiple chromatographic separation techniques, including silica gel,MCI gel and Sephadex LH-20 gel, were employed to isolate and purify the compounds. Their structures were identified by means of the nuclear magnetic resonance (NMR),mass spectrometry (MS) and physicochemical properties. Constituents were quickly analyzed by UPLC-LTQ-Orbitrap-MSn,and scanned in positive and negative ion modes. Result:These compounds were determined as trans-p-hydroxycinnamic acid(1),vanillic acid(2),protocatechuic acid(3),fumalic acid(4),succinic acid(5),succinic acid monomethyl ester(6),docosanoic acid(7),azelaic acid(8) and p-hydroxybenzaldehyde(9). Forty compounds were speculated by comparing mass spectrometry data,retention times of some compounds and reference materials,including 14 phenols and 26 organic acids. Among the compounds of herbal pair,8 phenols and 22 organic acids in Sparganii Rhizoma as well as 11 phenols and 13 organic acids in Curcumae Rhizoma were identified. Cleavage pathways of main compounds were described. Conclusion:There are abundant phenols and organic acids in Curcumae Rhizoma and Sparganii Rhizoma herbal pair and single herb. The results enrich pharmacodynamic material basis of Curcumae Rhizoma and Sparganii Rhizoma herbal pair.

OBJECTIVE:To sepa rate and identify the phenols compounds in Aurantii Fructus Immaturus ,and to provide reference for basic research of its effective substances. METHODS :The phenols compounds were isolated and separated by silica gel,HW-40F gel column ,ODS reversed column and preparation HPLC. Their physicochemical properties and spectral data were used to identify the structures. RESULTS & CONCLUSIONS :Totally 12 compounds were isolated from Aurantii Fructus Immaturus,identified as 6′-O-trans- cinnamoyl- 3,5-dihydroxyphenyl β-D-glucopyranoside(compound 1),methyl 3-(2′,4′- dihydroxy phenyl )propionate(compound 2),phloroglucinol(compound 3),aurantiside A (compound 4),5,7,4′-trihydroxy-8, 3′-dimethoxyflavone-3-O-6″-(3-hydroxyl-3-methylglutaroyl) β-D-glucopyranoside (compound 5), aromadendrin-7-O-β-D- glucopyranside(compound 6),hesperidin-7-O-β-D-glucoside(compound 7),naringin(compound 8),hesperidin(compound 9), narirutin (compound 10),neoeriocitrin (compound 11),eriocitrin (compound 12). Among them ,compound 1 is a new compound,and compounds 2 and 3 are isolated from Critus for the first time.

Abstract The aim of this work was to evaluate the effect of Salicylic acid, 2,4-D and 2-iP on the production of total phenolic and flavonoids as well in the levels of fukugetin and 7-epiclusianone in callus of Garcinia brasiliensis zygotic embryos. For this, Bacupari callus were exposed to different concentrations (0.0; 0.1; 1; 10; 100 µM) of Salicylic acid (SA) in the presence or in the absence of 72 μM 2-iP and 28.73 μM 2,4-D. The highest concentration of total phenolic occurred in the treatment with 100 μM SA in callus subcultured in the absence of 2,4-D and 2-iP, and with 10 μM SA in callus subcultured in the presence of these regulators. Concerning flavonoids, 2,4-D and 2-iP supplementation without the presence of the SA was sufficient for the highest levels. Additionally, it was possible to identify the fukugetin and 7-epiclusianone. However, the variation in the levels was very high, especially for the fukugetin. Therefore, in most treatments, there was no statistically difference. Except for the treatment with 10 μM SA + 2,4-D and 2-iP, where there was a significant increase in 7-epiclusianone.

Objective: To study the effects of salicylic acid (SA) and methyl jasmonate (MeJA) on growth, activity of related enzymes and chemical components in Polygala callus. Methods: The callus of Polygala was used as material. After 30 d of dark culture at different concentrations of SA (0, 4, 8, 12, 16, 20, 24, 28, 32 mg/L) and MeJA (0, 100, 200, 400, 600, 800, 1 000 μmol/L), the growth of callus, anti-oxidant enzyme activity, the content of MDA, total phenolic, total flavonoid, polygalaxanthone III, and 3,6’-disinapoyl sucrose were determined. Results: MeJA inhibited the growth of Polygala callus, and 12 mg/L SA promoted the growth of Polygala callus. SA and MeJA promoted the activity of SOD, CAT, POD, and MDA in the callus of Polygala. With the increase of SA and MeJA concentration, the activities of SOD, CAT and POD increased first and then decreased, and the content of MDA continued to rise. When the concentration of SA was 20 mg/L, the activities of CAT and SOD reached the maximum, which were 248.45 U/mg and 4451.06 U/mg, respectively. When the concentration of SA was 16 mg/L, the activity of POD reached the maximum, which was 7.22 U/mg. When the concentration of SA was 32 mg/L, the MDA content reached the maximum value of 25.09 nmol/mg. When the concentration of MeJA was 600 μmol/L, the activities of CAT, SOD, and POD reached the maximum, which were 273.30, 1451.06 and 15.27 U/mg, respectively. When the concentration of MeJA was 1000 μmol/L, the MDA content reached the maximum value of 27.10 nmol/mg. SA inhibited the accumulation of total flavonoids in Polygala callus, and had no significant effect on total phenols. MeJA promoted the accumulation of total phenols and total flavonoids in Polygala callus. When MeJA concentration was 600 μmol/L, the content of total flavonoids was the highest. When the concentration of MeJA was 400 μmol/L, the total phenolic content was the highest. Both SA and MeJA promoted the accumulation of polygalaxanthone III and 3,6’-disinapoyl sucrose in Polygala callus. When the concentration of SA was 32 mg/L, the concentration of polygalaxanthone III, 3,6’-disinapoyl sucrose was the highest. When the concentration of MeJA was 1 000 μmol/L, the content of polygalaxanthone III, 3,6’-disinapoyl sucrose was the highest. Conclusion: MeJA had an inhibitory effect on the growth of Polygala callus, and 12 mg/L SA can promote this effect. SA and MeJA promoted the activity of SOD, POD, CAT, the content of MDA and the accumulation of polygalaxanthone III and 3,6’-disinapoyl sucrose in Polygala callus. SA inhibited the accumulation of total flavonoids in Polygala callus, and had no significant effect on total phenolics. MeJA promoted the accumulation of total phenolics and total flavonoids in Polygala callus.
